Total Town Replacement Set modifications

I recently added the TTRS to OpenTTD, and I'm impressed by the high standard of the artwork.
I note in the readme and on the creator's website (http://users.skynet.be/florisjan/ttd/ttrs.html) that 4 parameters can be modified.
Sorry for the 'newbie' question, but how and where do I type newgrf/ttrs3[w].grf and how would the parameters be added to this? (eg would I type newgrf/ttrs3[w]0021.grf, for example?)

For anyone else reading the wiki, note that multiple parameters must be separated by spaces (I didn't notice on first glance).
