Can someone help me to get starting to create an Esperanto town names GRF file?

Sure. The first thing you need to do is make a list of town names (or parts of town names) you want to use. Turning the list into a grf is the relatively easy part.
Bone. La unua farendaĵo estas krei liston de la nomoj de urbetoj (aŭ de partoj de la nomoj de urbetoj), kiujn vi volas uzi. Ŝanĝi la liston en GRF-dosieron estas la plej facila parto.

OK, I just made a list with big cities in the world in Esperanto spelling, at least that can be a good start...

But seriously, I guess city names ending in o is an Esperanto thing? Like how lots of Greek last names end in s (in the Latin alphabet anyhow)?

The suffixes -o, -a, -e, and -i indicate that a word is a noun, adjective, adverb, and infinitive verb, respectively.
In Esperanto, all singular nouns end in "o," regardless of gender, and all plural nouns end in "oj." Nouns are turned feminine by the insertion of "in" between the word root and the "o" or "oj" suffix. For example:
* Hundo (dog)
* Hundoj (dogs)
* Hundino (she-dog)
* Hundinoj (she-dogs)

Well, to be honest I picked out just the completely esperantized ones, thus I discarded everything not finishing on o... a lot of cities are just used "as-is" as well

only very little, the basic grf info (grf-id, name, etc.) and a lang/default.lng file with the strings for english language. the details are explained somewhere on that page.
there should also be some examples on the dev zone already, e.g. german town names with a section of simple real city lists, and one section with building fictional names out of parts

If you install nml, it will generate a grf for you.

I, like the rest of the Esperantist OTTD population (approximately 3 total,) would like this GRF a lot. After seeing this thread, I created a list of Esperanto town names with two parts: 30 adjectives (like hela), and 31 nouns (like fonto.)
(EDIT: I'm working on turning this into a GRF, once I can wrap my head around it!)
